Overview

KMU-11361 is a novel small molecule multi-protein kinase inhibitor derived from indolin-2-one, currently under preclinical investigation for the treatment of rheumatoid arthritis (RA). Developed by researchers at Keimyung University, the compound demonstrates potent anti-inflammatory and anti-rheumatic effects by targeting multiple signaling pathways. It inhibits the phosphorylation of Transforming growth factor-beta-activated kinase 1 (TAK1), c-Jun N-terminal kinase (JNK), and the IKKα/β-IκBα-NF-κB axis. Furthermore, KMU-11361 suppresses the activation of the NLRP3 inflammasome and reduces the production of pro-inflammatory cytokines such as TNF-α, IL-1β, IL-6, and IL-2. In bone-related models, it inhibits RANKL-induced osteoclast differentiation by downregulating NFATc-1 and c-Fos, suggesting potential for preventing joint destruction in RA.
