KN-93 is a synthetic, cell-permeable small molecule and a selective inhibitor of calcium/calmodulin-dependent protein kinase II (CaMKII). By competitively blocking the calmodulin binding site of CaMKII, KN-93 inhibits enzyme activation and downstream phosphorylation events. It also acts as an extracellular open channel blocker of voltage-gated potassium channels (such as Kv1.5 and IKr), a property that is independent of CaMKII inhibition. KN-93 is used extensively as a research tool to study CaMKII-dependent signaling, neurobiology, and cardiac electrophysiology. It is not approved for clinical use and is intended for laboratory research purposes only[1][3][4][5][6][8][9].
