KN026 + docetaxel is a combination therapy consisting of KN026, a recombinant humanized bispecific antibody targeting the HER2 protein at domains II and IV (the binding sites for pertuzumab and trastuzumab), and docetaxel, a taxoid antineoplastic agent. This combination is being investigated as a first-line treatment for HER2-positive recurrent or metastatic breast cancer. KN026 blocks HER2 signaling by dual engagement of two distinct epitopes on the receptor, potentially overcoming resistance to standard anti-HER2 therapies. Docetaxel disrupts microtubule function during cell division, leading to apoptosis in rapidly dividing tumor cells. Clinical trials have demonstrated that this regimen achieves high objective response rates with manageable safety profiles in patients with HER2-positive metastatic breast cancer[1][3][5].
