Overview

KN026 + HB1801 is a combination investigational therapy currently in phase 3 clinical trials for the treatment of HER2-positive breast cancer. **KN026** is an anti-HER2 bispecific monoclonal antibody developed by Alphamab Oncology; it binds two non-overlapping HER2 epitopes (ECD2 and ECD4), resulting in dual blockade of the HER2 signaling pathway. **HB1801** is an albumin-bound docetaxel formulation (a taxane chemotherapy) designed for improved tolerability and effectiveness compared to traditional docetaxel. The combination aims to provide both targeted anti-HER2 activity (by KN026) and cytotoxic chemotherapy (by HB1801). This regimen is being studied as a novel first-line or neoadjuvant treatment for HER2-positive recurrent, metastatic, early, or locally advanced breast cancer, with the goal of achieving greater efficacy and safety versus current standard regimens involving trastuzumab, pertuzumab, and docetaxel[1][2][4][6][7].
