Overview

KN046 + lenvatinib is a novel investigational combination drug regimen for oncology consisting of KN046, a recombinant humanized bispecific antibody that targets both PD-L1 and CTLA-4, and lenvatinib, a small molecule multi-kinase inhibitor. KN046 works by simultaneously blocking the PD-L1 and CTLA-4 immune checkpoints, thus restoring and enhancing anti-tumor T-cell immune responses, while lenvatinib inhibits multiple receptor tyrosine kinases including VEGFR, FGFR, PDGFRα, RET, and KIT, targeting both angiogenesis and tumor proliferation. The combination is being explored as a first-line treatment in advanced unresectable or metastatic hepatocellular carcinoma (HCC) and has demonstrated promising efficacy and manageable safety in phase II trials[1][2][3][5]. KN046 is being developed by Jiangsu Alphamab, and lenvatinib is marketed by Eisai.
