Overview

KN056 is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist developed by Alphamab Oncology for the treatment of type 2 diabetes mellitus and obesity. It is a recombinant fusion protein consisting of a GLP-1 analogue fused to a human IgG Fc fragment, designed to extend its half-life through neonatal Fc receptor (FcRn) mediated recycling. This structural modification supports a once-weekly subcutaneous dosing regimen. In Phase 1 clinical trials, KN056 demonstrated dose-proportional pharmacokinetics with a half-life of approximately 141 to 188 hours and showed significant glucose-lowering effects and body weight reduction in healthy participants, while maintaining a favorable safety and tolerability profile.
