KN510 + KN713 + mFOLFIRINOX is an investigational combination therapy being developed by New Cancer Cure-Bio Co., Ltd. for the treatment of locally advanced or metastatic pancreatic ductal adenocarcinoma. The regimen combines KN510713—a proprietary dual-drug product consisting of two small molecule fatty acid oxidation (FAO) inhibitors, KN510 and KN713—with the standard-of-care chemotherapy backbone mFOLFIRINOX (modified fluorouracil, leucovorin, irinotecan, and oxaliplatin). KN510 acts as an inhibitor of the carnitine acylcarnitine carrier (CAC), while KN713 targets acetyl-CoA acyltransferase 1 (ACAA1). By simultaneously inhibiting these key components of the fatty acid oxidation pathway, the therapy is designed to disrupt the metabolic flexibility of cancer cells, thereby sensitizing them to the cytotoxic effects of chemotherapy. The combination is currently being evaluated in Phase I/IIa clinical trials to determine its safety and efficacy in patients with advanced pancreatic cancer.
