Overview

**KN510713** is an investigational oral small-molecule combination of **omeprazole** and **trimetazidine** being developed as a dual fatty acid oxidation inhibitor for cancer. Omeprazole, designated KN510 in this program, inhibits the mitochondrial carnitine-acylcarnitine carrier encoded by **SLC25A20**; trimetazidine, designated KN713, competitively inhibits **ACAA1**. Dual inhibition is intended to suppress fatty acid oxidation, ATP generation, autophagy flux, and downstream mTOR-associated acquired chemotherapy resistance. A completed Phase 1 study evaluated KN510713 in advanced solid tumors, and a Phase 1/2 study is evaluating KN510713 with modified FOLFIRINOX in locally advanced or metastatic pancreatic ductal adenocarcinoma.
