Overview

KND-1103 is an orally bioavailable small molecule inhibitor of dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A), currently in Phase 1 clinical development by KinoPharma. The DYRK1A gene is located within the Down Syndrome Critical Region (DSCR) on chromosome 21, and its overexpression due to trisomy 21 is a primary driver of cognitive deficits and early-onset Alzheimer's-like neurodegeneration in individuals with Down syndrome. KND-1103 is designed to normalize DYRK1A activity, thereby potentially improving synaptic plasticity and reducing the hyperphosphorylation of tau protein. Developed in collaboration with Kyoto University and Tokyo Medical and Dental University, the compound represents a more potent and selective therapeutic approach compared to earlier non-specific inhibitors like green tea extract (EGCG).
