KNP-301 is a bi-specific fragment crystallizable region (Fc) fusion protein designed to simultaneously inhibit complement component C3b and vascular endothelial growth factor (VEGF). This dual mechanism targets both the alternative pathway of the complement system and VEGF-mediated angiogenesis, aiming to treat retinal diseases such as late-stage age-related macular degeneration (AMD), including both neovascular (wet) and nonexudative forms. By blocking C3b, KNP-301 selectively inhibits the alternative pathway without affecting the classical pathway, while its VEGF trap function impedes pathological angiogenesis. Preclinical studies have demonstrated efficacy in animal models of AMD, with potential advantages including less frequent dosing due to a long intravitreal half-life. The drug is being developed by Kanaph Therapeutics for retinal and autoimmune diseases[1][2][4][5][6].
