Overview

KNX-115 is a potent, selective small-molecule inhibitor of Plasmodium falciparum myosin A (PfMyoA) actin-activated ATPase, with a Ki of approximately 17 nM and over 50-fold selectivity against vertebrate cardiac, skeletal, and smooth muscle myosins. Developed by Kainomyx in collaboration with TropIQ Health Sciences, KNX-115 acts as a multi-stage antiparasitic agent that traps PfMyoA in a state of low affinity for actin, thereby blocking parasite motility and host-cell invasion. It has demonstrated robust in vitro and ex vivo activity against multiple stages of the Plasmodium life cycle, including the asexual blood stage, liver stage, and mosquito-stage transmission, and remains effective against drug-resistant strains. Additionally, due to the evolutionary conservation of myosin A, KNX-115 exhibits broad-spectrum activity against other apicomplexan parasites, including Cryptosporidium, Toxoplasma, and Eimeria.
