Drug intelligence / Profile preview
KO7 CAR T cells (KO7CAR) is a second-generation, fratricide-resistant chimeric antigen receptor (CAR) T-cell therapy targeting CD7, developed by Hebei Senlang Biotechnology and Lu Daopei Hospital. To prevent fratricide (self-killing) among CAR-T cells, which naturally express CD7, the cells are modified using CRISPR/Cas9 gene editing to knock out (ablate) the CD7 gene prior to transduction with the anti-CD7 CAR construct. The CAR construct contains a second-generation design with 4-1BB (CD137) and CD3ζ co-stimulatory domains. KO7 CAR T cells are being evaluated in Phase I clinical trials for the treatment of relapsed or refractory CD7-positive hematological malignancies, including T-cell acute lymphoblastic leukemia (T-ALL), T-cell lymphoblastic lymphoma (T-LBL), and mixed phenotype acute leukemia (MPAL).
