KO7CAR is an experimental anti-CD7 chimeric antigen receptor (CAR) T-cell therapy developed by Hebei Senlang Biotechnology for the treatment of T-cell malignancies, such as T-cell acute lymphoblastic leukemia (T-ALL). A significant challenge in developing anti-CD7 CAR-T cells is fratricide, where the CAR-T cells target and kill each other because they naturally express the CD7 antigen. To overcome this, KO7CAR utilizes CRISPR/Cas9 gene editing to ablate (knock out) the endogenous CD7 gene in bulk T cells prior to lentiviral transduction with the anti-CD7 CAR construct. The CAR cassette typically includes a CD7-specific single-chain variable fragment (scFv) fused to a CD8 transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. In comparative studies, KO7CAR has been used as a benchmark to evaluate the performance of other approaches, such as naturally selected anti-CD7 CAR-T cells (NS7CAR).
