Overview

KOR-CRISPR-Cas9 is an experimental gene-editing tool designed to knock out the kappa opioid receptor (KOR) gene using the CRISPR-Cas9 system. In research settings, it has been employed to investigate the role of hypothalamic KOR signaling in migraine pathophysiology. By deleting KOR in the arcuate nucleus, researchers demonstrated a prevention of stress-induced prolactin release and subsequent trigeminal nociceptor sensitization in female mice. This tool is primarily used for functional genomics and target validation in the study of stress-induced pain and neurological disorders. It represents a targeted genetic approach to inhibiting KOR signaling permanently in specific brain regions.
