KOS-1584 is a second-generation synthetic epothilone with potential antineoplastic (anticancer) activity. It acts as a microtubule-stabilizing agent by binding to tubulin and inducing microtubule polymerization, which leads to rapid G2/M cell cycle arrest and apoptosis in cancer cells. Compared to earlier epothilones such as Epothilone D, KOS-1584 demonstrates higher potency (3–12 fold), improved pharmacokinetic properties including enhanced tumor tissue penetration and reduced central nervous system exposure, and favorable tolerability. It was developed primarily for the treatment of advanced solid tumors—including non-small cell lung cancer—and has shown activity in various tumor types resistant to taxanes[2][3][4][5][6][7][8].
