Overview

KOS-2484 is a second-generation epothilone analog that was under development by Kosan Biosciences as an antineoplastic agent. As a microtubule-stabilizing agent, KOS-2484 binds to the tubulin subunits of microtubules, promoting their assembly and preventing depolymerization. This mechanism results in the stabilization of the mitotic spindle, leading to cell cycle arrest at the G2/M phase and subsequent apoptosis of rapidly dividing cancer cells. KOS-2484 was designed to offer improved potency and a better safety profile compared to first-generation epothilones. Development of the compound was discontinued following the acquisition of Kosan Biosciences by Bristol Myers Squibb in 2008.
