KOSN-1724, also known as KOS-1724 or SK-10106, is a totally synthetic third-generation epothilone developed by Kosan Biosciences in collaboration with Roche. It functions by stabilizing microtubules, a mechanism akin to paclitaxel, thereby disrupting cell division. Preclinical studies have demonstrated its potent antitumor activity against various cancer cell lines, including leukemia, human lung carcinoma (A549), colon carcinoma (HCT 116), and mammary carcinoma (MX-1). In vivo, it achieved complete tumor remissions in nude mice bearing MX-1 and human ovarian adenocarcinoma (SK-OV-3) xenografts. The compound exhibits favorable stability in human plasma and liver microsomal fractions, along with significantly higher water solubility compared to paclitaxel. It has been investigated for both intravenous and oral administration.
