Overview

KPI-190 is a peptide-based therapeutic candidate derived from the ShK toxin platform, originally isolated from the venom of the sea anemone *Stichodactyla helianthus*. Developed by KPI Therapeutics, it was designed as a potent and selective blocker of the voltage-gated Kv1.3 potassium channel. This channel is a key regulator of effector memory T cells, which are implicated in the pathogenesis of various autoimmune and inflammatory conditions. KPI-190 was specifically formulated as a topical eye drop for the treatment of ocular inflammatory diseases, including uveitis and dry eye syndrome. Despite demonstrating proof-of-concept efficacy in models of anterior uveitis and showing good penetration into the anterior chamber when administered topically, development of the drug has been discontinued.
