KPT-127 is an early-stage small molecule selective inhibitor of nuclear export (SINE) developed by Karyopharm Therapeutics. It functions by irreversibly binding to and inhibiting Exportin-1 (CRM1), a nuclear export protein that is often overexpressed in various cancers, including pancreatic cancer. By blocking CRM1-mediated export, KPT-127 forces the nuclear retention and accumulation of tumor suppressor proteins (TSPs) such as p53, p27, and PAR-4. This localization restores the natural apoptotic pathways in malignant cells, leading to selective cancer cell death. Preclinical data presented at AACR 2012 indicated that KPT-127 and its analogs effectively inhibited growth and induced apoptosis in pancreatic cancer cell lines with minimal toxicity to normal cells.
