KQ-L6-PTA is an autologous, armored chimeric antigen receptor (CAR) T-cell therapy targeting Transmembrane 4 L6 family member 1 (TM4SF1). Developed by Shanghai Keqi Pharmaceutical Technology (and its subsidiary Shanghai Ultra-T Immune Therapeutics) in collaboration with the First Affiliated Hospital of Zhengzhou University, the therapy is engineered to secrete a bispecific immunomodulatory fusion protein called PTA. PTA consists of a PD-1 antibody and a TGF-beta inhibitor, designed to simultaneously block the PD-1/PD-L1 pathway and neutralize TGF-beta within the immunosuppressive tumor microenvironment. By combining direct TM4SF1-targeted tumor cell killing with localized immune checkpoint blockade and TGF-beta inhibition, KQ-L6-PTA aims to overcome microenvironmental resistance and enhance antitumor efficacy in patients with TM4SF1-positive advanced or recurrent solid tumors, including hepatocellular carcinoma and colorectal cancer.
