Drug intelligence / Profile preview
KQTD-126 is a potential first-in-class, gut-restricted pan-tropomyosin receptor kinase (pan-TRK) inhibitor developed by XtalPi. It is designed to treat chronic intestinal pain associated with irritable bowel syndrome (IBS) and inflammatory bowel disease (IBD). By targeting TRKA, TRKB, and TRKC receptors in the gastrointestinal tract, the drug regulates nerve activity and pain-signal amplification. KQTD-126 is engineered to be gut-restricted, with preclinical studies showing a gut tissue-to-blood exposure ratio exceeding 1,000:1 and sub-nanomolar potency (IC50 < 1 nM). This localized action is intended to confine its pharmacological activity to the gut, thereby avoiding the neurological side effects typically associated with systemic TRK inhibitors.
