Overview

KR23335 is a small molecule potassium channel opener developed by Shanghai Qingrun Pharmaceutical Technology for the treatment of epilepsy. It acts as a positive allosteric modulator of the KCNQ2/3 (Kv7.2/7.3) voltage-gated potassium channels, which are responsible for the M-current that regulates neuronal firing thresholds. By enhancing the activity of these channels, KR23335 promotes potassium efflux and stabilizes the resting membrane potential, thereby preventing the repetitive, high-frequency neuronal discharges characteristic of seizures. The candidate is currently in Phase 1 clinical development, aiming to provide a targeted therapy for patients with focal seizures and potentially those with KCNQ2-related developmental and epileptic encephalopathy (KCNQ2-DEE).
