**KRAS G12D-directed TCR-T cell therapy** is a target-specific, genetically engineered autologous T-cell therapy platform rather than one uniquely identified drug product. Patient T cells are modified to express a tumor-reactive T-cell receptor that recognizes a peptide containing the oncogenic **KRAS G12D** mutation when presented by a compatible human leukocyte antigen molecule on tumor cells. The engineered T cells can then recognize and kill KRAS G12D-positive cancer cells. Individual programs differ materially in their TCR sequence, HLA restriction, cell-manufacturing process, conditioning regimen, and eligible tumor types; publicly described examples include clinical programs for advanced solid tumors and gastrointestinal malignancies.
