KRAS-G12D-specific TCR-T cells are an investigational adoptive cell therapy (ACT) modality consisting of autologous T cells engineered to express T-cell receptors (TCRs) that specifically recognize the KRAS G12D neoantigen. KRAS G12D is a common oncogenic mutation in solid tumors, particularly pancreatic, colorectal, and non-small cell lung cancers. These engineered TCRs recognize the mutant peptide (e.g., VVVGADGVGK) presented by specific Human Leukocyte Antigen (HLA) molecules, such as HLA-A*11:01 or HLA-C*08:02. By targeting a neoantigen absent in healthy tissue, this approach aims to provide high tumor specificity and potent anti-tumor activity. Various academic institutions and companies, including Hangzhou Mechanodynamic Biotechnology, are developing specific clones (e.g., KDA11-01) using advanced engineering platforms to optimize binding affinity and reduce off-target toxicity.
