KRAS G12V CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy being developed by Myrio Therapeutics. It is designed to target the KRAS G12V mutation, a prevalent oncogenic driver in various solid tumors, including non-small cell lung cancer, pancreatic cancer, and colorectal cancer. The therapy utilizes antibodies discovered through the proprietary Retained Display (ReD™) platform that are engineered into CAR-T cells to recognize the intracellular KRAS G12V mutant peptide when presented on the cell surface by HLA-A*11:01 MHC complexes. This approach allows the immune system to target a traditionally "undruggable" intracellular protein by exploiting the natural MHC presentation pathway. The program is currently in the discovery stage, with preclinical evidence demonstrating efficacy in xenograft models of metastatic lung and pancreatic cancers.
