KRAS G12V-specific TCR refers to a T-cell receptor (TCR) engineered T-cell therapy (TCR-T) designed to target the G12V mutation in the KRAS oncogene. This shared neoantigen is a frequent driver in solid tumors such as pancreatic, colorectal, and non-small cell lung cancers. The therapy involves modifying a patient's T cells to express a specific TCR that recognizes the KRAS G12V peptide presented by Human Leukocyte Antigen (HLA) molecules on the surface of tumor cells. A prominent candidate in this category was developed by T-knife Therapeutics using their proprietary HuTCR humanized mouse platform, which identified a high-affinity TCR restricted to the HLA-A*11:01 allele. This approach aims to overcome the limitations of previous TCR-T therapies by providing optimal affinity and high specificity for the mutant epitope while avoiding off-target toxicity.
