KRAS G12V-targeted TCR-T cell therapy is a preclinical-stage adoptive cell therapy being developed by Fundamenta Therapeutics for the treatment of solid tumors. The therapy consists of autologous or engineered T cells that have been genetically modified to express a specific T-cell receptor (TCR) capable of recognizing the KRAS G12V neoantigen. This recognition is restricted to the HLA-A*11:01 allele, meaning the therapy is designed for patients carrying this specific human leukocyte antigen type. KRAS G12V is a prevalent oncogenic mutation found in various high-unmet-need malignancies, including pancreatic, colorectal, and lung cancers. By targeting this mutation, the TCR-T cells are engineered to selectively identify and eliminate tumor cells while sparing healthy tissue that lacks the mutant protein.
