The KRAS inhibitors developed by the Leuven Centre for Drug Design and Discovery (CD3) are small molecule compounds designed to target the KRAS protein, a member of the RAS GTPase family that is frequently mutated in various cancers. KRAS mutations, particularly at codons 12, 13, and 61, lead to constitutive activation of downstream signaling pathways such as MAPK/ERK and PI3K/AKT, driving uncontrolled cell proliferation and survival. These inhibitors aim to block KRAS activity, potentially treating a wide range of solid tumors where KRAS mutations are prevalent, including pancreatic, colorectal, and lung cancers. The project is currently in the preclinical development stage and is part of CD3's oncology pipeline, which focuses on translating academic research into novel therapeutic candidates.
