Overview

KRAS-specific autologous TCR-T cells are a form of adoptive cell therapy in which a patient's own (autologous) T lymphocytes are genetically engineered to express a transgenic T-cell receptor (TCR) that specifically recognizes neoantigens derived from mutated KRAS proteins presented by certain human leukocyte antigen (HLA) molecules on tumor cells. These therapies target common oncogenic mutations in the KRAS gene—such as G12D and G12V—which drive tumor growth in several solid cancers including pancreatic ductal adenocarcinoma and other advanced/metastatic solid tumors. The engineered T-cells are designed to recognize and eliminate cancer cells harboring these specific mutations. Some products, such as AFNT-211, also incorporate additional modifications like CD8α/β coreceptors for enhanced MHC-I recognition by CD4+ helper T-cells and FAS-41BB switch receptors to improve anti-tumor activity and prevent exhaustion[1][2][5][6]. This approach is being investigated primarily for patients with advanced or metastatic solid tumors carrying specific activating mutations in the KRAS gene.
