KRAS TCR-Transduced PBL is an investigational adoptive cell therapy developed by the National Cancer Institute (NCI). It consists of autologous peripheral blood lymphocytes (PBLs) that are genetically engineered via viral transduction to express murine T-cell receptors (TCRs) specific for oncogenic KRAS mutations, particularly the G12D and G12V hotspot variants. These TCRs are restricted by the HLA-A*11:01 allele, meaning the therapy is designed for patients who carry this specific HLA type and whose tumors harbor the targeted KRAS mutations. By reintroducing these engineered T cells into the patient, the therapy aims to induce a targeted immune response against metastatic or unresectable solid tumors, including pancreatic and colorectal cancers. It is currently being evaluated in Phase I and Phase II clinical trials, sometimes in combination with cancer vaccines to enhance the persistence and activity of the infused cells.
