A combination immunotherapy consisting of a synthetic long peptide (SLP) vaccine targeting common mutant forms of the KRAS protein (including G12D, G12R, G12V, G12A, G12C, and G13D) and the adjuvant poly-ICLC (Hiltonol). The SLP mKRASvax is designed to stimulate an immune response specifically against tumor cells harboring these KRAS mutations. Poly-ICLC is a synthetic double-stranded RNA that acts as an immune adjuvant by activating innate immunity through toll-like receptor 3 (TLR3), enhancing antigen presentation and T cell responses. This combination aims to induce robust anti-tumor T cell activity in patients with high-risk or advanced cancers driven by mutant KRAS. It is being investigated primarily for pancreatic cancer and other solid tumors with relevant mutations[1][2][4][5].
