Overview

KRIBB11 is a **synthetic small molecule** initially identified as a **selective inhibitor of Heat Shock Factor 1 (HSF1)**, the key transcription factor regulating cellular stress responses. By inhibiting HSF1, KRIBB11 suppresses the expression of heat shock proteins (notably HSP70 and HSP27), mitigating the development of chemoresistance in cancer cells and causing cell cycle arrest at the G2/M phase and apoptotic cell death. Preclinical studies have demonstrated its ability to reduce tumor growth in xenograft models without significant systemic toxicity. Beyond oncology, KRIBB11 also impacts the central nervous system: it acts on microglia via the **Akt pathway**, promoting process elongation, reducing neuroinflammation, inhibiting inducible nitric oxide synthase (iNOS) and NO production, and reversing behavioral abnormalities in mouse models of neuroinflammation.
