Overview

KRN5500 is a semisynthetic derivative of the nucleoside-like antineoplastic antibiotic spicamycin, originally isolated from the bacterium Streptomyces alanosinicus. It acts by inhibiting protein synthesis through interference with endoplasmic reticulum and Golgi apparatus functions, and also induces cell differentiation and caspase-dependent apoptosis. Initially developed as an anticancer agent with broad-spectrum activity against various human cancer cell lines, it has also been investigated for the treatment of chronic chemotherapy-induced peripheral neuropathy (CCIPN) in cancer patients refractory to conventional analgesics. Clinical studies have shown that it can provide relief from neuropathic pain in advanced cancer patients but is associated with gastrointestinal adverse events[1][3][5][6][8].
