KROS301 is a tumor-targeting small molecule and checkpoint inhibitor developed by Kairos Pharma. It functions by blocking the intranuclear localization of RelA (p65), a critical component of the NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) signaling pathway. By inhibiting the nuclear translocation of RelA, KROS301 aims to suppress cancer cell growth and mitigate immune suppression within the tumor microenvironment. The drug is specifically designed to target RelA/p65 biomarker-positive solid tumors, with a primary focus on advanced cancers such as triple-negative breast cancer (TNBC). Additionally, Kairos Pharma holds patents for its application in autoimmune diseases. As of early 2024, KROS301 is in the preclinical stage of development.
