Overview

KRP-A218 + itraconazole is a drug combination used in clinical pharmacology studies to evaluate the impact of CYP3A4 inhibition on the pharmacokinetics of KRP-A218. KRP-A218 (also known as HAR-1234) is a novel, orally active small molecule developed by Kyorin Pharmaceutical that acts as an ATP-competitive and subtype-specific inhibitor of phosphatidylinositol 4-kinase beta (PI4KB). By targeting this host cell factor, KRP-A218 inhibits the replication of RNA viruses such as human rhinovirus (HRV) and enteroviruses (EV). Itraconazole, a potent antifungal agent, is utilized in this combination as a strong CYP3A4 inhibitor to assess potential drug-drug interactions during Phase I clinical development.
