Overview

KS100 is a novel pan-aldehyde dehydrogenase (ALDH) inhibitor being investigated for the treatment of multiple myeloma (MM). It specifically targets both bulk MM cells and multiple myeloma stem-like cells (MMSLCs), which are characterized by a CD138-/ALDH+ phenotype and are associated with drug resistance and relapse. By inhibiting ALDH, KS100 prevents the detoxification of cytotoxic aldehydes and disrupts retinoic acid-dependent stemness pathways. Preclinical studies have shown that KS100 reduces clonogenic growth, exhausts the ALDH-high cell compartment, and demonstrates synergistic effects when combined with standard MM therapies such as proteasome inhibitors and immunomodulatory drugs (IMiDs), even in treatment-resistant models.
