KS99 is a synthetic isatin analog, specifically 5,7-dibromo-N-(p-thiocyanomethylbenzyl)isatin, developed by researchers at Pennsylvania State University for the treatment of various hematological and solid malignancies. It acts as a multi-target inhibitor, targeting Bruton tyrosine kinase (BTK), AKT, and tubulin polymerization. In preclinical studies, KS99 has demonstrated potent cytotoxic activity against acute myeloid leukemia (AML) cell lines and primary patient samples by inducing apoptosis through the modulation of Mcl-1 and Bcl-2 expression. It has also shown efficacy against multiple myeloma and several solid tumor models.
