KSP-QRH-E3-IRDye800 is a heterodimeric peptide-based imaging agent designed for the detection of cancers expressing both epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2). The molecule consists of two heptapeptide monomers—KSPNPRF (KSP) and QRHKPRE (QRH)—linked together and labeled with the near-infrared fluorescent dye IRDye800CW. This dual-targeting approach enables specific binding to EGFR and HER2 on tumor cells. The conjugation with IRDye800CW allows for dual-modal in vivo fluorescence and photoacoustic imaging, providing high tumor-to-background contrast for improved diagnosis, staging, and surgical guidance in cancer management. Preclinical studies have demonstrated high specificity for tumors overexpressing EGFR/HER2 with rapid clearance from non-target tissues. Developed by the University of Michigan College of Engineering, it was evaluated primarily as an optical imaging probe in gastrointestinal malignancies[1][2][5].
