KSQ-004EX is an investigational autologous cell therapy consisting of tumor-infiltrating lymphocytes (TILs) that are genetically engineered using CRISPR/Cas9 to inactivate both the SOCS1 and Regnase-1 genes. This dual-edit approach is designed to enhance the anti-tumor activity, persistence, and engraftment of TILs in patients with advanced solid tumors. Preclinical studies have shown that inactivation of these genes improves T cell function, tumor infiltration, resistance to exhaustion, and long-term memory formation. KSQ-004EX is being developed by KSQ Therapeutics for use in a variety of solid tumors including melanoma, non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), colorectal cancer (CRC), pancreatic cancer, and cervical cancer[2][3][4][5][7].
