Overview

KSQ-005 is an investigational cell therapy being developed by KSQ Therapeutics for the treatment of solid tumors. It utilizes the company's proprietary eCAR (enhanced CAR) technology, which integrates chimeric antigen receptor (CAR) engineering with CRISPR/Cas9-mediated gene editing of tumor-infiltrating lymphocytes (eTILs). Specifically, KSQ-005 is designed to inactivate two key negative regulators of T cell function: SOCS1 (Suppressor of Cytokine Signaling 1) and Regnase-1 (Regulatory RNase 1). By knocking out these 'genetic brakes,' the therapy aims to enhance the anti-tumor potency, metabolic fitness, and persistence of the engineered cells within the immunosuppressive tumor microenvironment. As of early 2024, KSQ-005 is in the IND-enabling stage of development.
