Overview

KT-485 is a **first-in-class, orally available, selective, and potent small molecule degrader of interleukin-1 receptor-associated kinase 4 (IRAK4)**, developed as an immuno-inflammatory disease therapy. Distinct from conventional kinase inhibitors, KT-485 uses targeted protein degradation (PROTAC approach) to eliminate IRAK4, thereby abrogating both its kinase and scaffolding functions. It is designed to provide a broad anti-inflammatory effect for diseases like atopic dermatitis, asthma, hidradenitis suppurativa, inflammatory bowel diseases, and rheumatoid arthritis. KT-485 is a **second-generation candidate** with greater potency and selectivity, and an improved safety profile over the prior candidate KT-474. It is being co-developed by Kymera Therapeutics and Sanofi and is entering Phase 1 clinical trials in 2026[1][2][3][4][6][7][8].
