KT-A112 is an investigational adeno-associated virus (AAV)-mediated gene therapy being developed by Kriya Therapeutics for the treatment of type 1 and type 2 diabetes, designed as a one-time intramuscular injection that delivers expression cassettes for human insulin and human glucokinase to skeletal muscle to restore physiologic glucose-responsive insulin production and improve glycemic control.[1][2][4][7][9][11] Preclinical studies in streptozotocin-induced diabetic animals have shown dose-dependent skeletal muscle expression of insulin and glucokinase, reductions in fasting and fed blood glucose, marked HbA1c improvements, normalization of triglycerides, and reversal of diabetes without hypoglycemia or need for immunosuppression, supporting further development toward clinical evaluation.[1][2]
