Overview

KT112 is a novel, cancer-specific monoclonal antibody targeting the epidermal growth factor receptor (EGFR). It was identified using a phage display library derived from the peripheral blood mononuclear cells (PBMCs) of an esophageal squamous cell carcinoma (ESCC) patient who exhibited long-term survival despite recurrence. Unlike conventional EGFR antibodies such as cetuximab or panitumumab, KT112 selectively binds to EGFR expressed on cancer cells and does not react with normal esophageal cells or the recombinant extracellular domain of EGFR. This high tumor selectivity suggests it recognizes a cancer-specific conformation or post-translational modification of the receptor. KT112 exhibits potent antibody-dependent cellular cytotoxicity (ADCC) and demonstrates high internalization capacity, making it a promising candidate for development as an antibody-drug conjugate (ADC) for the treatment of esophageal cancer.
