Overview

KTI-218 is a preclinical-stage small molecule covalent inhibitor developed by Kovina Therapeutics for the treatment of HPV-positive cancers and infections. It specifically targets the HPV-16 E6 oncoprotein by forming a covalent bond with cysteine 51 (Cys51). This binding disrupts the interaction between E6 and the E6-associated protein (E6AP), which is the ubiquitin ligase responsible for the degradation of the p53 tumor suppressor. By preventing E6-mediated p53 degradation, KTI-218 restores p53 levels and function, leading to apoptosis in HPV-transformed cells. The drug is being investigated for indications including HPV-positive cervical cancer, oropharyngeal cancer, and premalignant cervical infections.
