KTX-1001 + dexamethasone is an **investigational combination therapy** consisting of KTX-1001, a first-in-class, potent, oral, small molecule inhibitor of the histone methyltransferase MMSET/NSD2, combined with the corticosteroid dexamethasone. KTX-1001 acts by selectively inhibiting the catalytic activity of MMSET/NSD2, reducing the epigenetic mark H3K36me2, and thereby restoring balance to gene expression, particularly in multiple myeloma with the high-risk t(4;14) translocation. Dexamethasone is a standard-of-care synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressive properties, commonly used as part of myeloma regimens. The combination is under clinical investigation for relapsed and refractory multiple myeloma, including high-risk t(4;14) disease. KTX-1001 is developed and being investigated by K36 Therapeutics.
