Kuano is developing a first-in-class small-molecule inhibitor of NOTUM for the treatment of colorectal cancer (CRC), including treatment-resistant forms. NOTUM is an enzyme (carboxylesterase) that negatively regulates the Wnt signaling pathway by deacylating Wnt ligands; this pathway is frequently dysregulated in CRC to drive tumor growth and therapy resistance. By inhibiting NOTUM, the drug candidate aims to modulate Wnt signaling and restore its normal function to overcome treatment resistance. The lead molecules were designed using Kuano's proprietary quantum simulation and artificial intelligence platform and have demonstrated in vivo efficacy in mouse models. As of 2024, the program is in preclinical development with planned progression to organoid-based studies.
