Overview

KUR-111 is a bioactive bone graft substitute being developed by Kuros Biosciences as an in situ bone regeneration product for the treatment of fractures requiring grafting, particularly tibial plateau fractures. It is a combination of three components: a natural healing matrix based on fibrin sealant, a potent targeted bone anabolic agent (TGplPTH1‑34, a variant of human parathyroid hormone 1‑34), and a structural ceramic scaffold, which together form an injectable paste that polymerizes in situ to fill fracture voids, resist compression, and support bone ingrowth.[1][4][13][17] By covalently incorporating the PTH variant into the fibrin matrix, KUR-111 provides localized, controlled release of a parathyroid hormone receptor agonist to stimulate osteoblast-lineage cells and promote new bone formation, aiming to match or surpass autologous bone graft in efficacy while avoiding autograft-related morbidity.[3][4][7][17] In a large randomized Phase 2b trial in patients with tibial plateau fractures, KUR-111 met its primary endpoint by demonstrating non-inferiority to autograft in bone healing, with an excellent safety profile, and has been positioned as a potential new standard of care in severe bone trauma.[4][11][14][15][17]
