Overview

**KUR-503** is an off-the-shelf allogeneic chimeric antigen receptor (CAR)-modified natural killer T-cell (NKT) therapy developed by Athenex (following acquisition of Kuur Therapeutics) that targets **glypican-3 (GPC3)**, a cell surface protein overexpressed in hepatocellular carcinoma (HCC). It engineers NKT cells with a GPC3-targeting CAR, potentially enhanced by BATF3 overexpression for improved proliferation and tumor control, as shown in preclinical data presented at ASCO 2022 demonstrating superior survival versus IL-15 CAR-NKT cells in models. Designed for advanced solid tumors like **GPC3-expressing HCC**, it offers an allogeneic alternative to autologous CAR-T with reduced risks of cytokine release syndrome, neurotoxicity, or graft-versus-host disease.[1][2][3][5]
