Drug intelligence / Profile preview
KV998314 is a potent, selective, and orally bioavailable small molecule inhibitor of activated Factor XII (FXIIa) developed by KalVista Pharmaceuticals. It is designed to target the catalytic domain of FXIIa, blocking both the initiation and amplification of the intrinsic coagulation cascade and the kallikrein-kinin system (KKS). In preclinical models, KV998314 has demonstrated sub-nanomolar potency against both human and mouse FXIIa activity, with greater than 1000-fold selectivity over other serine proteases. It has shown efficacy in protecting against FeCl3-induced arterial thrombosis and inhibiting thrombin generation in human and mouse plasma without affecting hemostasis. The compound is being investigated for the prevention of thrombosis and hereditary angioedema (HAE) prophylaxis.
