KVA12123 is a human engineered IgG1 monoclonal antibody designed as a differentiated VISTA blocking immunotherapy to address immunosuppression in the tumor microenvironment (TME). It targets VISTA (V-domain Ig suppressor of T cell activation), an immune checkpoint molecule that negatively regulates T cell function and maintains immune quiescence. By blocking VISTA, KVA12123 aims to restore anti-tumor immunity, showing strong monotherapy tumor growth inhibition in preclinical models without evidence of cytokine release syndrome (CRS) in clinical trials. It is being developed primarily for advanced solid tumors including non-small cell lung cancer, colorectal cancer, renal cell carcinoma, head and neck cancer, and ovarian cancer. The drug is administered by intravenous infusion every two weeks and is being evaluated both as monotherapy and in combination with pembrolizumab (an anti-PD-1 therapy). The mechanism of action is complementary to other T-cell focused therapies.
